In total there are 57 story missions in Red Dead Redemption. They are listed below in the order you'll probably complete them, although in some cases you are able to choose a slightly different route.
Chapter 1: New Austin
There are 25 story missions in New Austin.
John Marston
John Marston arrives in Blackwater before being sent to Armadillo to track down an old friend.
Bonnie MacFarlane
John Marston recovers from his wounds before helping Bonnie MacFarlane clear out some vermin around her ranch.
Bonnie and John take part in a friendly horse race around MacFarlane's Ranch.
John and Bonnie take a wagon to the town of Armadillo to purchase some medicine from the Doctor's Office.
Bonnie needs help to guide some cattle from their pen out to pasture by an old oak tree. Give her a hand and start earning some cash.
John learns how to catch and break horses for MacFarlane's Ranch.
A storm is rolling in and Bonnie needs your help getting the cattle back to the ranch. Get the herd home safely before they become hamburgers.
Leigh Johnson
John Marston seeks help from the marshal in Armadillo. The two strike a deal to help each other out.
Cattle rustlers have been spotted in Pike's Basin so the marshal enlists the help of John Marston to help bring them down.
Nigel West Dickens
Escort Nigel West Dickens to a doctor in Armadillo before he bleeds to death. Along the way you'll need to protect him from some bandits.
Leigh Johnson
Assist Marshal Johnson with some murderous bandits terrorizing the countryside. It turns out their leader is an old friend.
Bonnie MacFarlane
Drew MacFarlane is missing and Bonnie is worried. She and John find her father alive at a bloody scene but while they're gone the ranch heats up.
Leigh Johnson
Bonnie has been kidnapped and it's up to John and the marshals to get her back before time - and Bonnie's oxygen - runs out.
Nigel West Dickens
Nigel West Dickens is out of the doctor's office and back to selling his elixir. He needs John's help to push his product to new customers.
Seth Briars
Help Seth get the other half of his map back from a former partner. You'll need to steal the partner away from some deputies, though.
Drive Seth to a secluded location while he checks some deceased friends for his treasure map.
Assist Seth with getting his treasure from the Tumbleweed mansion. The other treasure hunters won't make it easy, though.
Nigel West Dickens
Nigel needs to raise some more cash for the assault on Fort Mercer. A race is easy money for him, especially when he won't be doing the hard work.
Irish
Save Irish and follow him to the supposed location of a machine gun. Take out the guards around a cabin to get inside.
Once again Irish claims he's found a machine gun - this time it's in a mine. Fight through miners all the way to the bottom and claim the prize.
John has himself a gun but now he needs ammunition. Irish has a friend who can help with that - if he didn't get himself c-c-caught first.
Nigel West Dickens
Nigel continues to seek John's help to raise cash. He wants to sell some elixir one more time, but his previous customers catch up to him.
Once again Nigel is not quite ready to execute the planned assault on Fort Mercer. He needs just a little more cash and John's riding skills are proven.
Gun? Check. Ammo? Check. Friends to help? Check. Time to storm Fort Mercer and get Bill Williamson... if he hasn't fled to Mexico yet.
Irish
Marston gives chase as Bill Williamson flees deep into Mexico. Only problem entering the country is more "friends" of Irish.
Chapter 2: Nuevo Paraiso
There are 16 story missions in Nuevo Paraiso.
Vicente de Santa
Fresh blood in Mexico, Marston seeks help finding Escuella and Williamson from Captain de Santa but John will need to help the Mexican army before they will help him.
John meets Colonel Allende who says he will help John find Escuella and Williamson but first he needs some help fighting off rebels and sending them a fiery message.
Rebels have taken over the abandoned fort at Torquemada and Marston needs to help the Mexican Army clear them out. The rebels won't go easily, though.
Help a Mexican Army supply train through rebel-held territory. You probably aren't meant to come out alive, though.
Landon Rickets
Make friends with an old hero and learn a thing or two about shooting.
Landon heads out to rescue a part of his extended Mexican family with John coming along for support.
Face opponents in poker and then a duel when things go bad.
Take control of Mexican Army prisoner wagons with Landon and then take the prisoners to freedom in the United States.
Luisa Fortuna
Luisa needs her sister to flee the approaching revolution and get to safety elsewhere.
Rescue Abraham Reyes - the leader of the rebels - from the grip of the Mexican Army and bring him to safety.
Vicente de Santa
Colonel Allende says that Williamson and Escuella have been captured and are being held in Chuparosa... or so he says.
Luisa Fortuna
Luisa pleads for assistance in taking out an army supply convoy. Dynamite should work just fine.
Luisa wants De Santa dead. He may also know where Escuella is too. Don't expect De Santa to surrender easily.
Abraham Reyes
The rebels need supplies so Reyes wants to take some from the Mexican Army by stealing a train. They won't let it go easily, though.
Abraham Reyes finally finds out where Javier Escuella is hiding and takes John to him. You'll need to get through an army, though.
Time to finish off Allende. As one would expect, he won't be surrendering easily and may be harboring someone of interest.
Chapter 3: West Elizabeth
There are 7 story missions in West Elizabeth.
Edgar Ross
Assist Ross and Fordham infiltrating Dutch's supposed hideout at an old riverboat. Unfortunately the gang is smarter than some think.
Dutch van der Linde tries to rob the bank in Blackwater but the law intervenes. The fight gets taken across the region into the forest.
Harold MacDougal
Nastas knows where Dutch and his gang are hiding so he leads the way in an effort to infiltrate the enemy base.
Escort MacDougal and Nastas to meet with some of Dutch's Native men. Apparently talking is not actually something they want to do.
Help MacDougal escape Blackwater from Dutch and his savages. Snipers and cavalry won't make it easy, though.
Edgar Ross
Ross wants to make a final assault to kill Dutch van der Linde with the help of the US Army. Of course Dutch won't give up without a fight.
After wrecking the army's new toy, it's time to go back to horses in order to put a bullet in Dutch... if he allows it.
Chapter 4: Beecher's Hope
There are 9 story missions in Beecher's Hope.
Abigail Marston
After a long and troublesome adventure it's finally time to see the Marston family again. The ranch has some assembly required.
Damn crows keep stealing corn. A scarecrow made of lead will probably scare them to death.
The MacFarlanes lost all of their corn sacks and desperately need supplies. Luckily the Marstons have plenty to give.
Uncle
Now that the Marstons have cows, the cattle need get out for food and a healthy run - perhaps more than needed or expected.
Wild mustangs are spotted roaming the plains. They're worth a good amount so might as well capture a few.
Jack Marston
It's time for Jack to learn hunting. Elk should be easy targets and provide some valuable items.
Wolves have been bothering the Marston cattle herd. Time to step up Jack's hunting lesson.
Jack decides to go hunting on his own. Not the best idea for someone in his position.
Just when the Marstons' lives were getting back to normal the past has one card left to play. The name of the game is sacrifice.
